Phylogenomics and evolution of Superrosids Kates, Heather [1], Folk, Ryan [2], Guralnick, Robert [3], Soltis, Pamela [4], Soltis, Douglas [1]. Building and using a massive-scale 13,000 species phylogeny for the nitrogen-fixing clade. One single clade contains all flowering plant lineages known to undergo nodule development with bacterial symbionts that fix nitrogen. About half of the more than 30,000 species in the nitrogen-fixing clade are thought to form this symbiosis. These “nitrogen-fixing species” occur across 10 families in four orders (Rosales, Cucurbitales, Fagales, Fabales). A clear understanding of the evolutionary history of this critical symbiosis is needed to uncover the genetic basis of the nodulation trait and to ask how nitrogen-fixing symbioses within the nodulating clade contributed to the diversification of angiosperms. Because of the broad but variable distribution of the nodulation trait across the hugely diverse nitrogen-fixing clade, robustly reconstructing the evolutionary history of nodulation requires a densely sampled phylogeny inferred from a phylogenomic dataset. Here we present the largest phylogenetic estimate yet for the nitrogen-fixing clade: a phylogeny of over 13,000 species inferred from 94 nuclear loci. We include representative sampling of the diversity of all major clades in the tree through massive sampling of herbarium specimens and thus enable a greater focus on the non-legume origins of N-fixing symbioses compared with previous studies. We consider recent tests of the hypothesis that a single predisposition for nodulation was followed by multiple gains and losses of the symbiosis using our robust phylogeny, which includes major reorganizations in key taxa, and a database of the nodulation trait that has been updated based on recent literature, especially in the non-legume families
