Preliminary annotation and analysis of the Arctostaphylos glauca genome.

Arctostaphylos (Ericaceae), also referred to as manzanita, is a diverse woody genus that is composed of over 100 morphologically-defined species and subspecies, 75% of which are endemic to the Californian Floristic Province (CFP), its center of diversity. This genus is well known for complicated patterns of interspecific and intraspecific morphological variation, due to at least partly to hybridization and morphological overlap among species. Currently, there is not enough informative genetic data to test species delimitations. We are doing a large scale study using reduced-representation sequencing to investigate genetic diversity within this genus. To facilitate the detection of genetic variants, we used Oxford nanopore sequencing and short-read Illumina sequence data to produce a de novo assembled genome of Arctostaphylos glauca, which is a widespread and ecologically important species in the CFP. Here, we report the first chromosome-scale genome assembly and annotation of this genus and conduct a preliminary analysis in the context of other Ericaceae and Ericales. With the predicted gene sequences, we explore the history of whole genome duplication in A. glauca, syntenic changes compared to other sequenced Ericaceae genomes, and differentiation in gene content across the family.
